哮喘模型-屋尘螨引起的哮喘

背景简介：

哮喘是由接触尘螨、宠物皮屑、花粉或霉菌等过敏原引起的，其特征是气道受阻和支气管痉挛。在这些过敏原中，屋尘螨（HDM）是人类最重要的室内过敏原。两种主要的螨虫种类，屋尘螨（D.pteronyssinus）和粉尘螨（D.farinae）是导致人类过敏源性哮喘的主要变应原。

Chondrex提供的屋尘螨和粉尘螨纯化提取物，非常适合诱导小鼠过敏性哮喘，用于构建与人类过敏性哮喘非常相似的小鼠哮喘动物模型。此外，Chondrex的室内尘螨抗体检测试剂盒、细胞因子检测试剂盒和趋化因子检测试剂盒，也可用于监测HDM诱导的哮喘模型的进程，并测定实验疗法的效果。其Der p10检测试剂盒和胰蛋白酶活性检测试剂盒，还可用于测定过敏原含量和总蛋白酶活性水平。

 

1）小鼠过敏性哮喘模型

哮喘新疗法的研究需要相应的动物模型，尽可能的再现人类哮喘的情形：气道高反应性（AHR）、可逆性气道阻塞、早期和晚期哮喘反应、气道重塑和嗜酸性粒细胞增多症。传统方法使用卵清蛋白（OVA）诱导小鼠过敏性哮喘。然而，OVA不是临床上的过敏原，铝佐剂的体内致敏过程与人类由过敏原致敏的过程截然不同。因此，研究人员转向使用临床相关的过敏原，如屋尘螨HDM。HDM是一种免疫复杂的过敏原，可以刺激先天性和适应性免疫反应。与传统的OVA诱导哮喘模型相比，HDM诱导的过敏性哮喘更接近于人类过敏性哮喘。HDM诱导的过敏性哮喘模型实验疗法可能会为哮喘患者治疗提供新的选择。

 

2）屋尘螨（House Dust Mite, HDM,）

HDM提取物，主要来自于屋尘螨和粉尘螨，是人类最重要的室内过敏原。它包含20种顺序和功能同源的过敏原。其中，Der 1和Der 2是诱导患者免疫反应的主要过敏原，也是研究中最常见的过敏原。Der p 1和Der f 1是半胱氨酸蛋白酶，可以切割紧密连接并增加气道上皮的通透性。这种蛋白水解提升了抗原细胞识别过敏原的可能性，从而诱导促炎反应。哮喘诱导尤其与Der p1蛋白水解活性相关，可以用胰蛋白酶测定法用显色底物进行测量。

 

Der p 2和Der f 2与MD2的氨基酸序列具有高度同源性，MD2是脂多糖（LPS）的共配体，通过TLR4信号激活先天免疫反应。除了Der 1和Der 2，HDM还含有其他HDM过敏原，以及细菌和真菌产物，如LPS和β-葡聚糖。因此，HDM可以在不使用佐剂的情况下激发适应性和先天性免疫反应。与需要铝佐剂的OVA诱导的哮喘模型不同，HDM提取物可单独用于诱导过敏性哮喘实验。

 

实验流程：

1）屋尘螨诱发哮喘时间点

目前已发表多种小鼠诱导HDM哮喘模型的实验方案。图1-3显示了不同剂量HDM下，过敏原激发过程的示例方案。我们建议根据您的研究需要建立和优化方案，因为HDM的有效剂量取决于小鼠的品系、饲养条件和HDM的质量。此外，长期接触HDM会导致气道重塑，粘液细胞密度和AHR增加，即使在停止接触HDM后，AHR也会保持升高。

2）哮喘模型评估

屋尘螨诱发哮喘模型可以再现人类过敏性哮喘的情形。但其疾病表型可能因研究条件而异，如小鼠品系、HDM质量和挑战方案。因此，如何选择疾病评估方法，对于评估疾病严重程度和治疗效果尤为重要。

a.小鼠抗屋尘螨抗体分析

建立HDM诱导的哮喘模型需要高浓度的HDM IgE抗体。与传统OVA模型和人类哮喘相比，HDM模型中的IgE反应往往较弱。其中，HDM-IgG1抗体与Th2应答相关，而HDM-IgG2a或IgG2b与Th1应答相关。因此，测量HDM IgE和IgG的抗体浓度有助于判定致敏类型并实施对应的治疗方案。Chondrex的小鼠尘螨抗体测定试剂盒和小鼠总免疫球蛋白测定试剂盒，可以量化小鼠样品中HDM抗体的特定亚型和亚类。

b.细胞因子和趋化因子分析

c.BALF中炎性细胞计数与特定气道阻力

在HDM诱导的哮喘模型中，诱导哮喘会增加支气管肺泡灌洗液（BALF）中中性粒细胞、嗜酸性粒细胞、淋巴细胞和巨噬细胞的免疫细胞计数。此外，气道高反应性（AHR）在疾病进程中也会随之加重。
